Summary of key aims
ITALIAN MINISTRY OF HEALTH "RICERCA CORRENTE" - Line 5
The project "Apoptosis and cutaneous diseases", developed by the Laboratory of Skin Biochemistry, focused on the analysis of the mechanisms underlying epidermal differentiation and programmed cell death of keratinocytes. We have investigated the role of Nitric Oxide (NO), free radicals, and lipoxigenases in the induction of apoptosis of epidermal keratinocytes. We have demonstrated that NO is responsible for S-nitrosylation of cysteine proteases, such as caspases and transglutaminases, involved in apoptosis. Since the UV light is one of the apoptotic stimuli to which the skin is normally exposed, the effects of UV irradiation in keratinocytes were also evaluated.
In parallel, the role of the transcription factors of the p53 family in cell death and differentiation of keratinocytes has been investigated. Both the p53 homologues, p73 and p63 are expressed in epidermal keratinocytes and are able to regulate DNA damage-induced apoptosis, by either the death receptor- (CD95, TRAIL-R), or the mitochondria-mediated apoptotic pathways.
The projects "Oxidative stress and strategies to counteract skin ageing" and "Relationship between systemic and cutaneous ageing", developed by the Laboratory of Cell Ageing, focused on cutaneous aging and specifically on
ƒ{ Evaluation of systemic and cutaneous oxidative stress in relation to age, sex, UV exposure and photoprotection;
ƒ{ Evaluation of skin immune response after antioxidant oral treatments;
ƒ{ Evaluation of efficacy of sun screen and/or free radicals scavengers in inhibiting UV induced skin damage.
The project "Molecular and cellular mechanisms of skin ageing, as a basis for the optimization of dermatological rejuvenation procedures", developed by the Laboratory of Tissue Engineering and Cutaneous Physiopathology, is focused on the study of molecular markers and biochemical pathways involved in the senescence of skin cells and extra-cellular matrix. Free radical production /antioxidant functional status of the individual patient, specific for age related modifications, is of main interest. On the basis of results obtained, the optimisation of clinical protocols for the treatment of age related skin defects and rejuvenation is ongoing.
ITALIAN MINISTRY OF HEALTH "RICERCA FINALIZZATA 2002"
The project ¡§Health care quality in older dermatological patients: evidence based treatment, quality of life and satisfaction¡¨ developed by the Laboratory of Health Services focused about hospitalised patients > 65 with psoriasis and leg ulcers hospitalized. With validated instrument we tested 326 patients (161 and 165), showing the use of Evident Based Treatment in 80% and 73% of patients affected by psoriasis and leg ulcers, respectively.
